If you are concerned with voltage spacing, check out the PCB design threads on this forum. There is a range of conventions that builders follow depending on how conservative they are.
Generally, if you are using traditional hand assembly methods, there will be no concern about voltage creepage and arcing if you make room for your own hands to work in there.
You drills will last longer if you invest in a real drill press rather than a press that holds a drill, although even that is a huge improvement over holding the drill by hand. With a drill press you will find that HSS can drill hundreds or even thousands of holes in fiberglass card stock for eyelets.
